Surface Plasmon Resonance (SPR) is a physical phenomenon in which linearly polarized light, under conditions of total internal reflection, undergoes resonant energy transfer at a metal-dielectric interface. This principle enables the label-free analysis of molecular interactions. Ligand molecules are immobilized on a metallic film (typically gold), followed by the injection of analytes (e.g., proteins, DNA, or small molecules). When analytes bind to the ligand, they induce changes in the refractive index at the interface, resulting in a corresponding shift in the SPR angle. By monitoring these changes in real time, SPR provides dynamic interaction data between biomolecules. This technology is widely applied in biomolecular interaction studies and drug discovery, offering unparalleled advantages over conventional methods.

Affinity Analysis of Biosimilars
Using a Protein A chip, a rabbit anti-human FGL1 monoclonal antibody (developed by Yurogen) was captured. The analyte, recombinant human FGL1 protein with a His tag (ABclonal, Cat: RP01268LQ), was introduced at varying concentrations. The interaction was analyzed using the Polariton Life S-CLASS system, and the equilibrium dissociation constant (KD) was determined to be 0.11 nM.

  • High-throughput screening using Protein A chips.
  • Test 1 or 2 analyte concentrations per sample pair.
  • Affinity (KD) and binding kinetics (Ka/Kd) data are obtained and sorted according to customer requirements.
  • Specificity analysis can be provided if control analytes are available.
  • One data point is defined as a single analyte at one concentration per sample pair.
  • A minimum of 10 sample pairs will be analyzed.
